Tightness:
Metal tightness via piston with cone shaped seat 
Radial soft seal with packing, o-ring etc. 
Sample Ball Valve with PTFE/Metal sealing up to 180°C

Actuation:
Hand wheel or lever depending on nominal diameter, spring loaded lever (dead-man's safety system) single or double acting pneumatic drive, (dependant on DN/PN and / or hub) electric or hydraulic drive, air pressure motor.
